An Efficient Protocol for Micro Propagation of Ginger (Zingiber Officinale Rosc.) cv. Volvo via Sprout Bud Culture

An efficient, simple micro propagation method was developed for Zingiber officinale Rosc.) cv. Volvo using fresh sprouting bud in basal culture media. Explants were cultured on to Murashige and Skoog’s (MS) medium supplemented with different concentrations combinations of BA (Benzyl adenine) KIN (Kinetin) shoot IBA (Indol-3-Butyric acid) NAA (α- Naphthalene acetic root induction. MS 2.0 mg/L + 0.5gm/L gave the highest averages number (11.3 shoots/explant), leaf (22.8 leaves/shoot) plant height (16.4 cm) followed by concentration 2 1mg/L (9.8 (13.9 (10.3 cm). In vitro shootlets rooted half strength media 1.0 2mg/L obtained roots length achieved (9.6 roots/shoot 11.1 cm respectively) (8.3 10.8 respectively. Rooted shoots transplanted green house hardening off planted at mixture forest soil, coffee husk sand soil (2:1:1) ratio respectively their survival 96% field condition. Hence, acclimatization seedlings set edible rhizomes cement bug pot seven months.
